ARA-290 is a synthetic 11-amino acid peptide derived from the B-helix region of erythropoietin (EPO), specifically engineered to activate tissue-protective signaling without stimulating erythropoiesis. It is a targeted investigational molecule used in studies of neuroinflammation, ischemia-reperfusion injury, and non-hematopoietic EPO receptor activity.

Unlike native EPO, ARA-290 does not engage the classical EPOR homodimer responsible for red blood cell production. Instead, it selectively binds to the Innate Repair Receptor (IRR)—a heterodimer composed of the EPOR and the common beta subunit (CD131). This receptor complex is activated during cellular stress, particularly in vascular, neural, and immune tissues, initiating downstream cytoprotective responses. ARA-290 has gained traction in preclinical models for its role in modulating inflammation, enhancing nerve function, and supporting tissue regeneration without altering hematocrit or stimulating hematopoietic progenitor cells.
